How are the Lipid-Soluble Antioxidants analyzed?
Genox incorporates internal standards into the sample prior to extraction. This minimizes errors due to sample loss or variations in injection volume. Our isocratic HPLC procedure provides simultaneous measurement of the carotenoids, tocopherols and retinol. Other features of Genox’s assay include:
· Sample |
100 mL serum (carotenoids, tocopherols and retinol) 200 mL serum (CoQ10) |
· Method |
Ethanolic protein denaturation followed by hexane extraction and HPLC analysis |
· Conditions |
Isocratic mobile phase and C18 reversed-phase column |
· Detection |
Diode array detection of retinol, carotenoids and CoQ10 Fluorescence detection of tocopherols |
· Quantitation |
Recovery and quantitation are based on the method of internal standards |
· Repeatability |
Serum extraction and instrument precision: RSD < 5% for all compounds. |
In order to verify the continued validity of our method, Genox analyzes a QC serum sample with each assay. Our QC sample has been standardized using the Standard Reference Material (SRM) for Fat-Soluble Vitamins, provided by the National Institute of Standards and Technology (NIST). Genox-determined values for the carotenoids, tocopherols and retinol agree closely with the NIST- certified values.
In addition, Genox participates in a NIST-sponsored analytical round robin studies frequently, where the carotenoids, tocopherols and retinol and quantitated in unknown sera. Results from over 60 laboratories are tabulated and graded. In all of the round robin studies participated by us, determinations provided by Genox were within the acceptable range for all analytes.
