1 Application to tissue samples CFE-010 can be applied to cell lysates and tissue homogenates.
Add 5% TCA solution. Please confirm that pH of the mixture is pH2 - 3.
Incubate for 30 minutes at 4-8 °C.
Centrifuge for 15 minutes at 6,000 rpm.
Collect the supernatant, and apply to CFE-010.

NOTE: Centrifuge should be performed after acid extraction.

1 Application to cultured cells CCU-400 can be applied to cultured media and cell homogenates. Please take care that chelating agents such as EDTA may interfere this assay.

[Assay example]
1) Add HCl at a final concentration of 0.1 M, and extract for 15min - overnight.
2) Centrifuge at 5,000 - 10,000 rpm for 15 min 4 °C
3) Collect the supernatant and apply to CCU-400 assay.
2 Aged frozen tissue Q: We have frozen tissue which has been frozen in liquid nitrogen for over 6 months.
Can we use this kind of sample?
A: Yes, but please avoid freeze / thaw cycles.

1 Application to plant materials
(Acid extraction)
Typical protocol for acid extraction:
1) Prepare plant homogenates.
2) Add hydrochloric acid (HCl) or Nitric acid at a final concentration of 0.01 - 0.1 mol/L.
3) Mix vigorously and stand for 1 hour.
4) Centrifuge the mixture at 5,000-10,000 rpm for 10 minutes at 4 degree C to remove insoluble materials.
5) Apply to Zn assay kit.

In the application plant materials, breakdown of cell walls would be needed for extraction process. Please take care to prepare homogenates into fragments small enough to effective extraction. Experimental conditions can be modified depending on the nature of samples.

1 Application to bone samples: 1) Wash bone sample with saline, desiccate it and measure weight.
2) Mix 0.1g of homogenated bone sample with 1mL of 0.1 M of HCl, and shake overnight.
3) Centrifuge 10,000 rpm for 10 min at 4°C, and collect supernatant.
4) Apply to Ca assay kit (dilution will be needed).

Experimental conditions such as extraction time, HCl concentration and volume should be optimized depending on your samples. If HCl with higher concentration will be used, please adjust pH between pH2 and 3 by NaOH solution.
2 Demineralized bone samples 1) Acid cleavage by nitric acid and hydrogen peroxide, heat at 200 °C.
2) Dissolve dried samples with 0.1M HNO3.
3) Adjust pH between pH2 and 3 by NaOH solution.
4) Apply to Ca assay kit (dilution will be needed).

2 Demineralized bone samples 1) Acid cleavage by nitric acid and hydrogen peroxide, heat at 200 °C.
2) Dissolve dried samples with 0.1M HNO3.
3) Adjust pH between pH2 and 3 by NaOH solution.
4) Apply to Ca assay kit (dilution will be needed).
3 Stool samples: 1) Homogenate stool sampls into powder or small particles.
2) Mix 0.1g of homogenated stool sample with 1mL of 0.1 M of HCl, and shake for 30 minutes.
